چکیده
PIM kinase is consequently emerging as a promising target for cancer therapeutics and immunomodulation. kinases are overexpressed in variety of hematological malignancies solid tumors, their inhibition has become strong therapeutic interest. Currently, some pan-PIM inhibitors being developed under different phases clinical trials. Based on the scaffold structures, they can be classified into various subclasses. The X-ray structure complex outlines rationale hit compound confirmation early stage. Structure–activity relationships allow us to rationally explore chemical space further optimize multiple physicochemical biological properties. This review focuses discovery development small-molecule current research, hopes provide guidance future exploration inhibitors.

Multivalent Small-Molecule Pan-RAS Inhibitors
Design of small molecules that disrupt protein-protein interactions, including the interaction of RAS proteins and their effectors, may provide chemical probes and therapeutic agents. We describe here the synthesis and testing of potential small-molecule pan-RAS ligands, which were designed to interact with adjacent sites on the surface of oncogenic KRAS. One compound, termed 3144, was found to...
متن کاملActivation of Pim Kinases Is Sufficient to Promote Resistance to MET Small-Molecule Inhibitors.
Mesenchymal-epithelial transition (MET) blockade offers a new targeted therapy particularly in those cancers with MET amplification. However, the efficacy and the duration of the response to MET inhibitors are limited by the emergence of drug resistance. Here, we report that resistance to small-molecule inhibitors of MET can arise from increased expression of the prosurvival Pim protein kinases...

Different animal models have been proposed to investigate the mechanisms of Human T-lymphotropic Virus (HTLV)-induced pathogenesis: rats, transgenic and NOD-SCID/γcnull (NOG) mice, rabbits, squirrel monkeys, baboons and macaques. These systems indeed provide useful information but have intrinsic limitations such as lack of disease relevance, species specificity or inadequate immune response. An...
